1.12 Å Resolution on Apoferritin: New Krios 5 Commissioned at NeCEN

Commissioning of the new Thermo Scientific Krios 5 (Cold-FEG 300 keV, Falcon 4i detector, Selectris filter) is complete at NeCEN in Leiden. An automated overnight collection on ApoF reached 1.12 Å resolution, demonstrating that the instrument is tuned and ready for user projects.

1.12 Å ApoF Benchmark

To test performance at high magnification, we collected an overnight single-particle dataset on apoferritin (ApoF). Standard processing in cryoSPARC yielded a reconstruction from 400,000 particles at 1.12 Šresolution (gold-standard FSC = 0.143 cutoff; B-factor of 20.3 Ų).

At 1.12 Å, aromatic ring holes and carbonyl oxygens are resolved, protrusions corresponding to hydrogen atoms are visible in the density, and high-threshold contouring isolates individual atomic peaks. The benchmark confirms that the new Krios 5 in Leiden is ready for user collections across both academic and industrial projects.

User Access

The Krios 5 is open to both academic and industrial researchers.
